Shoot organogenesis and plant regeneration from leaf and petiole explants of Corydalis saxicola Bunting

Abstract Corydalis saxicola Bunting, a medicinal herb known for alkaloids, grows mainly in Guizhou province and northwest Guangxi of China. In this study, the first time plant, an efficient adventitious shoot proliferation regeneration system via organogenesis from leaf petiole explants was established. Cytokinins, including 6-benzyladenine (BA) thidiazuron (TDZ), induced callus then organogenesis. Most shoots were on Murashige Skoog (MS) medium (pH 6.5) supplemented with 2.0 µM BA 0.5 α-naphthaleneacetic acid (NAA). This also optimal proliferation. The rooting ½MS 1.0 indole-3-butyric (IBA) 0.2 NAA; 95.7% roots within 30 d 91% plantlets survived peat:vermiculite (1:1, v:v) substrate after 90 d.
